Summary

Nitric oxide synthases (NOSs) are heme proteins that catalyze the formation of nitric oxide (NO) from L-arginine and oxygen in a sequential two-step process. Studies of the ligand-protein interactions in NOS will be discussed. Focus will be placed on the interactions of NO with the enzyme as it has been shown that this catalytic product auto-inhibits the enzyme. Spectroscopic studies will be presented that reveal a significant heme distortion in the enzyme and regulation of the monomer/dimer equilibrium by NO and the tetrahydrobiopterin cofactor.
