概 要

Microbial rhodopsins are photoactive retinal-binding membrane proteins performing ion-pumping and photosensory functions in archaea, bacteria, and lower eucaryotes. From numerous genomes and metagenomes sequenced in the last decade, it became clear that the family of microbial rhodopsins is much more diverse than was thought originally. This includes taxonomic, structural, functional, and ecological diversity, making microbial rhodopsins important players in the biosphere. While more than a thousand of microbial rhodopsin gene variants is known, just a few of them is characterized in-depth biophysically. Our group (in collaboration with several others) works on novel rhodopsins from fungi, algae, cyanobacteria, and proteobacteria, applying spectroscopic methods, such as time-resolved FTIR, solid-state NMR, and several others, to characterize novel structures and functions of these proteins. I will survey our studies of microbial rhodopsins, concentrating on overexpression and characterization of new fungal photosensors, as well as on structural studies of marine proteorhodopsin.
