概 要

Solid-state NMR is gradually emerging as a powerful technique in structural biology to obtain information about structure and dynamics for so-called insoluble proteins. These include membrane proteins, amyloid fibrils, and protein complexes being intrinsically difficult to study by X-ray diffraction and liquid-state NMR spectroscopy requiring the proteins to be either in well-ordered 3D crystals or being mobile in liquid environment. Solid-state NMR has a unique potential for structural analysis of proteins in native heterogeneous environments.

In this presentation, I will illustrate the application of NMR in the study of insoluble proteins through three different examples. The first will be the combined use of liquid-state NMR and oriented solid-state NMR to study the action of antimicrobial peptides through formation of ion channels and ways to alter/enhance such actions through chemical modification. Focus will be devoted to alamethicin and novicidin. The second theme will be the use of solid-state NMR supplemented by biophysics, AFM, and magnetic resonance imaging in studies of amyloid fibrils, their formation, structure, and interactions with small-molecule binders for diagnostic purposes. Focus will here be devoted to fragments of amylin, prions, and Ab. The third topic, demonstrated the use of solid-state in combination with liquid-state NMR and cryoEM, to study proteins in heterogeneous native environments, address studies of the antennal system in the chlorosomes of green sulfur bacteria.
