概 要

Channelrhodopsins (ChRs) are sensory microbial rhodopsins that have become renowned for being the first and so far the only light-gated ion channels known in nature. ChR2 from the alga Chlamydomonas reinhardtii has been used for nearly a decade to depolarize the cell membrane of excitable cells by light, making it a prominent tool in the field optogenetics. The functional mechanism of ChRs remains relatively poorly understood in molecular terms.
In our group we have used time-resolved optical and vibrational spectroscopies combined with site-directed mutagenesis to gain information about the molecular mechanism that links retinal isomerization to the opening of the ion channel. Here, I will provide an overview on proton transfer reactions occurring in the photocycle of ChR2, and the role they might play in the closure of the conductive state of the channel. I will also present new results on the role and nature of conformational changes accompanying the formation of the conductive state of the channel.
